No, Japanese is a Phonic Language.
Every character always has the exact same pronunciation which never changes. Unlike English that has about 1000 different Rules and 1000 different ways to break each of those rules, Japanese just has a strict policy: "Character 'A' is pronounced exactly This way, and no other way at all".
エ - Romanized "E" - Pronounced "Eh" like "Head" without the H or D.
ニ - Romanized "Ni" - Pronounced like "Knee"

ス - Romanized "Su" - Pronounced like the "Su" in "Suit"
When they're trying to render Western languages in Japanese, they sometimes end up with an extra vowels due to them not having any flat consonants other than "N", so when it's being rendered back into Western style, it's typical to drop those ONLY when transliterating into English (Like Usoppu -> Usopp). Oda writing "Enies Lobby" all over the place proves Oda intended this.
So we have "Eh-Knee-Ehz"
It's three Syllables, not two.
Everyone in this set is saying it "Eh-Kneez" and it's annoying… T_T
