I have a Wacom Intuos 2 and I think it's great (got it in a fantastic deal) but one day I hope to have one of Wacom's fabled LCD screen tablets
Answering your questions, tablets are basically mice in pen form, so asking what programs are compatible with tablets is the same as asking what programs are compatible with mice
Mice and tablets can be connected at the same time and be used one after the other on the fly, but can't be used simultaneously (once I thought my computer crashed when I couldn't move my mouse until I discovered my pointer was on top of my tablet)
Moving the pointer over the tablet (but not touching) moves the mouse and touching signifies a left click (leaving it touched signifies holding the click). There is a button on the pointer used for the right click
