My money is on the crew getting split up. But it could be anything since this was one epic year in OP.

The whole year was insane but essentially two moments really capped it:
1. Duval: the fact that he just literally turned that joke Wanted poster into a character had to be one of the funniest moments Oda has ever done.
2. The Straw Hats annihilated: Just when we figured we knew a bit of where the story was going, Oda pulled something so shocking with Kizaru, Sentoumaru and ultimately Kuma that he literally blew away all our expectations! Anything he does now is literally controlled by him. -
Flying fish raiders base looking like the trading post from waterworld.

I still think it was the total defeat and split-up of the crew. He totally changed the pace of the story without jumping any shark. That blew me away.
I agree, that's the biggest twist in the whole series.

1st utter defeat of the straw hat crew by kuma was the big surprise….
its obvious oda was pointing to there... he made them feel invincible at that point... they had just beat CP9, ran from a buster call, owned moria (2nd shicibukai), owned duval, hit a world noble... all pulling a false sense of invicibility of the protagonists...
and then bang... kuma pwns everybody... zoro bend down on his knees... crew running away, nami crying... luffy totally impotent.
to me that is a the surprise

Exactly how is the defeat of the entire crew shocking? especially when it already happened with Aokiji years ago?
Kuma beat the crew up after they were already worn out by Moria/Oz. Hell they were worn out when Kizaru came to pwn them as well. It was great but not a surprise. Oda doesn't frequently use the shounen plot device to bail out his protagonists. If they gonna lose it'll be in grand fashion, just so they return with something new to win with…......

All the year was great and everything was a surprise… When TB was finishing, I thought that the next place was fishman island and now we are at Impel Down trying to rescue ace with the help of buggy after the suddenly split up of the crew WTF?!
I think this was the best year of one piece, but if I have to say one moment that was a surprise is the split up of the straw hats

well it seems splitting of crew is obvious because who would have ever thought they would all be split up and on different islands.
i thought buggy was a neat surprise, but really looking at all we listed as being surprises ODA didn't even need to tell us what that big surprise is, we can all find our own big surprise pretty easy

for me:
1. luffy is son of dragon and garp is luffy's grandfather
2. aokiji says to robin, that ohara is not dead yet … it is more like a hope to robin
3. split of the crew? this is not a surprise, it is just another adventure of them, but the surprise is, kuma was saving them from kizaru ... that is the big surprise ...and other thing, after luffy eat that mushroom in little amazon, will that cause another side effect beside sprouting mushroom from his hole body? i really hope that he will have another upgrade of power after that ...
so he can do ... gomu gomu no super giant jet bazooka.

Somehow with the rate that Fishman Island has been delayed and hinted since first mentioned way back with Arlong, I'm starting to think that there won't be anything major there and it will more be like a massive break after all the heck that's going on with Impel Down and the WB war. (particularly if Jinbei gets treated as an ally instead of an enemy with recent revelations)

For me the biggest surprise and most memorable moment was the defeat of the crew. I remember reading the chapter like 10 times, more than any other chapter this year, because it was just awesome.
I even translated it for my girlfriend so she could read it. I almost had tears in my eyes the first time i saw the pictures. Great story-twist.I wonder what it will be like, when the members meet each other again after this event.

It has to be big since it should be where the whole slavery storyline comes to a head at.

Definitely the whole crew splitting up thing.
There were some great twists earlier in the year, too, but Oda said he wasn't sure if he'd get to what he was talking about in 2008.
Given that there were only a few months between then, there's no way he could have been talking about something like revealing Roger or Rayleigh. There was no way that the Duvall stuff would have lasted over a year.
What a twist it was, too. The story has been so different lately, and yet it still feels wonderful. Most authors can't pull that off, but Oda has.

And I certainly hope it is! But I'd be surprised as hell if it wasn't which would also be a good thing.
I really have no idea how Oda intends to handle this Tera-Arc but it's going to play a role somehow and kick ASS!

Well, since as I said I take this to be the Oda version of an Atlantis story (which of course always are about ancient civilizations) I'd be really surprised if we don't get more hints about the OP ancient civilization, the void century or about a new Ancient Weapon. To make it short, if there's no Poneglyph at Fishman Island I'll cry.=/
